Biuret forms a purple-red complex in an alkaline solution of copper sulfate and potassium sodium tartrate, and its absorbance is measured at a wavelength of 550 nm.
4 Reagents and solutions

6.1.1 Preparation of standard colorimetric solution
As shown in Table 1, inject the dipivoxel standard solution into eight 100 mL volumetric flasks in turn. Approved by the General Administration of Quality Supervision, Inspection and Quarantine of the People's Republic of China on July 26, 2001 and implemented on January 1, 2002

Each volumetric bottle is diluted with water to about 50 mL, and then 20.0 mL of potassium sodium tartrate alkaline solution and 20.0 mL of copper sulfate solution are added in sequence, shake well, dilute to the scale, and immerse the volumetric bottle in a water bath at 30℃±5℃ for about 20 minutes, shaking from time to time. 6.1.2 Absorbance determination
Within 30 minutes, use the solution with zero biuret as the reference solution, and use a spectrophotometer to measure the absorbance of the standard colorimetric solution at a wavelength of 550nm.

Use the mass (mg) of diaminobenzene contained in 100mL standard colorimetric solution as the horizontal axis and the corresponding absorbance as the vertical axis to draw a graph, or find the linear regression equation.
6.2 Determination
6.2.1 Preparation of test solution
According to the different contents of diaminobenzene in urea, determine the sample weight according to Table 2 and weigh the sample accurately to 0.002 g. Then carefully transfer the weighed sample to a 100mL volumetric flask, add a small amount of water to dissolve (the amount of water added should not exceed 50mL), place it at room temperature, add 20.0mL potassium sodium tartrate alkaline solution and 20.0mL copper sulfate solution in turn, shake well, dilute to the scale, and immerse the volumetric flask in a water bath at 30℃±5℃ for about 20 minutes, shaking from time to time.

6.2 Determination
6.2.1 Preparation of test solution
